While Jet recorded a demo of "Are You Gonna Be My Girl", the studio microphones accidentally picked up a cough during the introduction. The producers wanted it in the final mix, but Nic Cester refused to fake the cough. They copied it from the demo and edited it in.

Cester had written an early version between the ages of 17 and 19. The original lyrics were about being rejected by a girl, with the hook "She's just like every other girl". Cameron Muncey fleshed it out and convinced Cester to be more positive. The hook became "Are you gonna be my girl". The song is often compared to Iggy Pop's "Lust for Life", but the band says it owes more to 1960s Motown, and Chris Cester said the beat came from there.

It came out on August 18, 2003, in the United States as the first single from Get Born, and on September 1 in Australia. It reached number 29 on the Billboard Hot 100, number three on the Modern Rock Tracks chart, and number 20 in Australia, and it was voted number one in the 2003 Triple J Hottest 100. In the black and white video, filmed at Vinopolis in London, ink pours out of the band's equipment and forms a scene.
