The release of the "Reptilia" single slipped slightly after Julian Casablancas objected to the credit on its B-side, a duet called "Modern Girls & Old Fashion Men". It was billed as "the Strokes and Regina Spektor", and he claimed it should read "Regina Spektor and The Strokes".

The song came out on February 9, 2004, as the second single from the Strokes' second album, Room on Fire. Casablancas wrote it and Gordon Raphael produced it. In the US it peaked at number 19 on the Modern Rock Tracks chart. It did better in the UK, reaching number 17 and going gold with the British Phonographic Industry. The cover shows an alien from the video game Centipede.
